Rainbow Six Vegas 2 speelt zich parallel af met haar voorganger. Deze keer maak je echter deel uit van een ander team en speel je Bishop. Bishop is één van de Rainbow top bevelhebbers en een collega van Logan Keller, de held uit Rainbow Six Vegas 1. Ook dit nieuwe team moet de strijd aangaan met de terroristen van Alvarez Cabrero om de strijd voor eens en altijd te beslechten.
